Project subject – Mechanisms of gastric bypass-induced diabetes remission
Project Details:
The fact that GBP causes remission of T2D makes this group of patients suitable for identification of curative processes for T2D. We now focus on T2D- and GBP-induced expression changes in the small intestine.
It is not well studied how the intestinal mucosa is affected by GBP or if alterations in the gut play a role in remission of T2D in humans. Recent animal studies point towards a role for the jejunum (alimentary limb) in the remission of T2D. However, unbiased assessment of global gene expression is lacking. We perform transcript profiling, using RNAseq in jejunal specimens taken during surgery and from the same location one year postGBP during gastroscopy in obese diabetic subjects.
Pathway analysis suggests that certain unexpected biochemical processes are activated after GBP. We now aim to validate these changes at the protein level in patient tissue as well as in established animal models. To address functional roles we will use Villin-Cre mice to target genes of interest specifically in the gut. The mice will be subjected to glucose-tolerance tests, assessed for gut morphology (e.g. L-cell density and glucose-transporter expression/localization), and given 3H-glucose to measure intestinal glucose uptake. We will also use intestinal cell lines and siRNA or CRISPR silencing to address the function of hits.
Qualifications:
We are seeking a highly motivated person with a PhD (Degree 2014-2017) in a relevant area and at least one first authorship publication. Excellence in molecular biology and state of the art histology. Proven skills in bioinformatics is meriting. A strong background in the diabetes field is a merit but not necessary. Excellence in the English language (written and spoken) is a prerequisite.
Eligible candidates must have obtained their PhD within the last three years. Moreover, eligible candidates must not have held positions at Lund University within the last two years.
