The MAX DELBRÜCK CENTER FOR MOLECULAR MEDICINE (MDC) is a member of the Helmholtz Association of National Research Centers, supported by the Federal Government of Germany and the Land Berlin. For the Molecular Pathways in Cortical Development, we are looking as soon as possible for one

  Postdoctoral Scientist

  Description:

In the developing central nervous system (CNS) many signalling pathways work in concert to ensure that neural stem cells are appropriately directed to generate progenitor cells, which mature into neurons and glia. Precise regulation of morphogen and growth factor activity is central to normal fate decision of neuronal progenitors and early embryonic brain patterning. Many congenital malformations in patients including holoprosencephaly (HPE) and neural tube closure defects (NTDs) involve mutations in components of the sonic hedgehog (SHH) and WNTsignalling machineries. Data from animal models and human patients suffering fromHPE and NTDs has driven significant progress in elucidating the biogenesis and the intracellular downstream pathways of morphogens. However, the pathways at the level of signal recognition and intra- and intercellular trafficking of morphogens are still poorly characterized.

Our work focuses on the role of the endocytic receptors of the low density lipoprotein receptor related protein (LRP) family in modulating SHH, WNT signalling and folate uptake. In particular, we study the complex regulation of morphogen and growth factor activity at the level of signal recognition and interpretation on the surface of the target cells during neural development and in the adult cortical stem cell niche.

Overall we aim at elucidating molecular mechanisms that regulate the differentiation of neural precursor cells. We study these questions both in vivo using mouse models and in vitro using neural stem cell cultures and cephalic explant cultures.
