摘要:
In our lab we aim to understand how the spatial-temporal organization of the genome contributes to the regulation of nuclear processes. Currently, the main research topics in the lab focus on elucidating the role of chromatin and nuclear architecture in gene-regulation and DNA-repair. To account for heterogeneity between individual cells of a population and to enable the study of rare cell-types, we employ and develop techniques to study genome-organization in single-cells. We have developed a microscopy-based tool to monitor and live trace the dynamic genome-architecture (Kind et al Cell 2013) and more recently and very excitingly, we designed a method to profile DNA-protein interactions in single-cells (Kind et al in preparation). With these methods at hand, the objective of this project is to gain detailed insight in the role of chromatin and nuclear organization in re-programming the mouse genome during pre-implantation development. To this end, we plan to generate single-cell profiles of several key chromatin types and nuclear organization at different cell-stages of development. Statistical modelling of the resulting datasets will be applied with the ultimate goal to uncover the determinants of cellular re-programming during early embryogenesis.
